Tumor necrosis factor receptor superfamily member 10C is a protein that in humans is encoded by the TNFRSF10C gene. The protein encoded by this gene is a member of the TNF-receptor superfamily. This receptor contains an extracellular TRAIL-binding domain and a transmembrane domain, but no cytoplasmic death domain. This receptor is not capable of inducing apoptosis, and is thought to function as an antagonistic receptor that protects cells from TRAIL-induced apoptosis. This gene was found to be a p53-regulated DNA damage-inducible gene. The expression of this gene was detected in many normal tissues but not in most cancer cell lines, which may explain the specific sensitivity of cancer cells to the apoptosis-inducing activity of TRAIL. An upstream start codon, which has a weak Kozak signal and is conserved in some other primates, is also present. The use of the upstream start codon would result in a protein that is 40 aa longer at the N-terminus. However, this would eliminate a signal peptide that is predicted for the shorter but not the longer N-terminus. PMID:9242611 shows experimental evidence for cleavage of a signal peptide following aa 25 from the downstream start, and this is also confirmed in PMID:15340161.
