Sentrin-specific protease 1 is an enzyme that in humans is encoded by the SENP1 gene.
The covalent modification of proteins by the small ubiquitin (UBB; MIM 191339)-like protein SUMO (see SUMO1, MIM 601912) is implicated in the regulation of nucleocytoplasmic transport, genomic stability, gene transcription, and other processes. Sumoylation is catalyzed on target lysine residues by a multienzyme process and is reversed by desumoylating enzymes such as SENP1 (Yamaguchi et al., 2005 [PubMed 15923632]).[supplied by OMIM, Jul 2008]. Function: Protease that catalyzes two essential functions in the SUMO pathway: processing of full-length SUMO1, SUMO2 and SUMO3 to their mature forms and deconjugation of SUMO1, SUMO2 and SUMO3 from targeted proteins. Deconjugates SUMO1 from HIPK2. Deconjugates SUMO1 from HDAC1, which decreases its transcriptional repression activity.
